The Chromosomal Aberration Test of YH1885 in Chinese Hamster Ovary Cell In vitro

Abstract

YH1885 is one of 750 H+/K+-ATPase inhibitors produced by Yuhan Resarch Center and is a new candidate for therapy to stomach ulcer. To investigate the clastogenecity of YH1885, we performed chromosomal aberration assay with chinese hamster ovarian
cells
in vitro. The IC50 value of YH1885 was determined as 37.5§¶/ml in Neutral Red assay. The concentrations for the chromosome test were thus 37.5, 18.75, 9.37§¶/ml, respectively. No significant increase in chromosome aberrations was observed at any
of
these concentration both in absence and presence of metabolic activation system. Mitomycin C and cyclophophamide caused a significant increase in chromosome aberration.
Thus, it may be concluded that YH1885 is not capable of inducing chromosome aberration in cultured chinese ovarian cells without regards to metabolic activation and genotoxicity of that is negative under the present experimental condition.
